It's true that adding weight to an exercise is one way to produce more stimulus to muscles, but to answer the question: yes, muscles do get used to exercises. Adding weight is obviously one way of keeping your muscles from getting used to the exercise, or plateauing. There are other things you can do too. You can reduce rest time, add more reps, add more sets, use dumbbells if you usually use a barbell or vice versa, switch the set/ rep scheme (for example, instead of 4 sets of 8, 8 sets of 4), using a different but similar exercise, the list goes on. The important thing to remember is that after about a month of doing an exercise the same way, switch it up. This prevents you from plateauing.
